When embedded into a Tailed Beast, it could directly extract their chakra.
By using a specific technique, Jiraiya could then absorb the stolen chakra into his own body, fusing it with his own energy, and permanently claiming the power of a Tailed Beast as his own.
In the original timeline, Tailed Beasts couldn't truly die. They were formed from chakra, and even if slain, their chakra would eventually disperse into the world and reassemble over time, resurrecting them.
However, if their chakra was completely absorbed into another being and never returned to the world, then they would cease to exist entirely.
Jiraiya's sign-in reward allowed him to accomplish precisely this.
Shukaku's power was being drained away, and unlike before, this time, he wouldn't have the chance to revive.
